CLEAVAGE OF PLANT CHROMOSOMES BY RESTRICTION ENDONUCLEASES

Cytological observations were made on the root-tip cells of Hordeum vulgare and H. bulbosum treated with a site-specific endonuclease preparation (restriction enzymes Hind II and III from Haemophilus influenzae). The exogenously supplied endonuclease had cleaved the chromosomes in intact cells of both species into discrete fragments. Degradation was progressive with time. The number of breaks per H. vulgare chromosome was greater than per H. bulbosum chromosome. The possible implications of such differential susceptibility and practical applications of this technique are discussed.
