失血性休克致肠屏障改变大鼠回肠组织中紧密连接蛋白ZO-1与Occludin的表达

摘要
目的研究紧密连接蛋白ZO-1、Occludin在失血性休克(hemorrhagic shock)致肠屏障改变大鼠回肠组织中表达的变化。方法健康雄性SD大鼠30只,随机分为假手术组和实验组,每组15只。假手术组不制作休克模型;实验组制作失血性休克模型。观察两组大鼠造模后24h采用Chiu 6级评分法对肠黏膜损伤程度进行评价,观察回肠组织病理形态、超微结构变化;免疫组化分析两组大鼠造模后24h回肠组织上皮细胞紧密连接分子ZO-1、Occludin的分布和表达。ELISA法检测两组大鼠造模后24h血浆D-乳酸水平。结果假手术组大鼠全部存活,实验组有13只大鼠失血性休克肠屏障损伤造模成功;实验组大鼠肠黏膜损伤评分高于假手术组(P<0.05);透射电镜下观察假手术组大鼠细胞间紧密连接较长,实验组大鼠上皮细胞紧密连接增宽;实验组大鼠血浆D乳酸值高于假手术组(P<0.05);实验组大鼠ZO-1及Occludin平均灰度值均低于假手术组(P<0.05)。结论失血性休克致肠屏障改变可降低大鼠回肠组织紧密连接蛋白ZO-1与Occludin的表达。
