低频重复经颅磁刺激对卒中后抑郁大鼠行为学及海马c-fos、c-jun蛋白表达的影响

摘要
目的:探讨重复经颅磁刺激(rTMS)对卒中后抑郁(PSD)模型大鼠行为学及海马c-fos、c-jun蛋白表达的影响。方法:筛选40只健康雄性SD大鼠按随机数字表法分为对照组、假手术组、模型组和治疗组(n=10)。对照组不给予任何干预处理;模型组和治疗组采用大脑中动脉栓塞法(MCAO)和慢性不可预见温和应激法(CUMS)建立PSD模型;假手术组和治疗组给予rTMS治疗;通过蔗糖水消耗实验、旷野实验和水迷宫实验对各组大鼠行为学进行评估;应用免疫组化实验检测各组大鼠海马c-fos、c-jun的阳性细胞灰度值。结果:(1)蔗糖水消耗实验:与模型组比较,治疗组蔗糖水消耗量明显增多(P<0.05);(2)旷野实验:治疗组大鼠水平及垂直得分较模型组有显著提高(P<0.05);(3)水迷宫实验:治疗组大鼠较模型组学习和记忆能力都有所改善(P<0.05);(4)免疫组化实验:治疗组大鼠c-fos阳性细胞灰度值高于模型组(P<0.05),各组间c-jun阳性细胞灰度值差异不显著(P>0.05)。结论:rTMS能够有效改善PSD模型大鼠的抑郁样行为,其机制可能与海马c-fos、c-jun蛋白的表达有关。
