依达拉奉对Aβ1-40诱导PC12细胞NF-κB、Bcl-2mRNA和蛋白表达的影响

摘要
目的探讨依达拉奉对β淀粉样蛋白(Aβ1-40)诱导PC12细胞NF-κB、Bcl-2 mRNA和蛋白表达的影响。方法采用Western印迹法,RT-PCR等检测NF-κB、Bcl-2 mRNA和蛋白的表达,观察MCI-186对其保护作用。结果模型组中NF-κBP65 mRNA和蛋白表达水平明显升高,Bcl-2 mRNA和蛋白表达水平明显降低,与对照组比较有显著差异(P<0.01)。保护组中NF-κBP65 mRNA和蛋白及Bcl-2 mRNA和蛋白表达与模型组组间比较有统计学意义(均P<0.05),而与对照组比较无统计学意义(P>0.05)。结论Aβ1-40可以活化神经细胞内NF-κB,减少Bcl-2的表达,发生细胞凋亡。依达拉奉可以抑制NF-κB激活,增加Bcl-2的表达发挥抗凋亡作用,最终达到保护神经细胞的目的。
