ADH1B对鼻咽癌细胞的增殖及迁移能力的影响

摘要
目的:鼻咽癌(nasopharyngeal carcinoma,NPC)已成为我国头颈肿瘤患者的主要病死因素之一,局部治疗失败和远处转移是晚期NPC患者不良预后的主要原因。大量研究证明,ADH1B是多种癌症的风险基因,本研究探讨ADH1B在鼻咽癌细胞中的表达差异以及对鼻咽癌细胞增殖和迁移能力的影响。方法:利用生物信息学方法分析ADH1B在头颈鳞状细胞癌(head and neck squamous cell carcinoma,HNSC)与正常组织中的相对表达量,应用实时荧光定量PCR(qRT-PCR)检测正常永生化鼻咽上皮细胞NP69及鼻咽癌细胞CNE-1、CNE-2、5-8F、6-10B中ADH1B的mRNA表达水平,对比两种研究结果的一致性。在此基础上建立稳转ADH1B高表达鼻咽癌细胞系,以pCDH作为空载体对照。利用生长曲线实验检验鼻咽癌细胞的增殖能力,利用划痕实验检验鼻咽癌细胞的迁移能力。结果:生物信息学分析结果显示ADH1B在头颈鳞状细胞癌中表达量低于正常组织。qRT-PCR检测发现,ADH1B在鼻咽癌细胞CNE-1、CNE-2、5-8F、6-10B中均比NP69细胞表达低,过表达ADH1B组鼻咽癌细胞的生长曲线低于pCDH组(P<0.05),划痕愈合速度也低于正常表达组pCDH组(P<0.05)。结论:鼻咽癌细胞中ADH1B基因表达异常,明显低于正常鼻咽上皮细胞,与生物信息学分析结果一致。ADH1B基因在鼻咽癌中高表达影响鼻咽癌细胞的增殖和迁移,这与NPC的发生发展密切相关,可能是参与调控NPC发生发展的一个重要靶标。
