益气化瘀方缓解关节不稳型小鼠膝骨关节炎的实验研究

摘要
目的:观察益气化瘀方是否能够缓解关节不稳诱导的小鼠膝骨关节炎病理改变,初步探讨疗效机制。方法:取12周龄C57BL/6小鼠30只,分为假手术组、模型组和益气化瘀方组,模型组和益气化瘀方组左膝关节行内侧半月板及前交叉韧带离断术诱导膝骨关节炎,并分别給予生理盐水和益气化瘀方煎剂灌胃。8周后取左侧膝关节,Micro-CT扫描后进行三维重建、阿尔辛兰/橙黄G染色以及Ⅱ型胶原免疫组织化学染色,观察膝关节软骨病理改变。结果:术后8周,模型组小鼠左侧膝关节内侧半月板钙化明显增加,关节软骨边缘骨赘形成,关节软骨表面不平整,出现破裂,阿尔辛兰和Ⅱ型胶原染色层变薄;益气化瘀方组小鼠膝关节稍有软骨边缘骨赘形成和半月板钙化,但无模型组显著,关节软骨表面稍有不平整,未出现破裂,阿尔辛兰和Ⅱ型胶原染色层稍有变薄,但无模型组显著。结论:益气化瘀方可以减轻半月板和前交叉韧带离断诱导的小鼠膝骨关节炎病理改变。
