地黄寡糖改善HepG2细胞胰岛素抵抗的分子机制研究

摘要
目的研究地黄寡糖改善HepG2细胞胰岛素抵抗的分子机制。方法运用RT-PCR技术,检测地黄寡糖对胰岛素抵抗HepG2细胞内过氧化物酶体增殖物激活受体α(PPAR-α)、胰岛素受体(IR)、葡萄糖转运蛋白4(GLUT4)和2(GLUT2)基因表达的影响。结果地黄寡糖能够促进胰岛素抵抗HepG2细胞中PPAR-α、IR、GLUT4 mRNA的表达;能够明显降低GLUT2基因mRNA的表达。结论地黄寡糖对HepG2胰岛素抵抗具有明显的改善作用,其机制可能与激活PPAR-α、调节HepG2内IR及GLUT2 mRNA的表达有关。
