细胞骨架与机械信号传导:椎间盘突出机制研究的新靶点

摘要
椎间盘突出的起始病因不清。作用于椎间盘上的非正常机械力及其导致的细胞外基质代谢变化是椎间盘退行性病变的起始原因。细胞骨架蛋白如肌动蛋白、微管蛋白和波形蛋白是机械信号传导通路的重要组成部分,可以将胞外机械刺激信号转换为细胞生物学反应,以帮助维持椎间盘组织胞外基质成分的正常代谢和更新。多种因素尤其是年龄会导致椎间盘细胞中细胞骨架蛋白结构和表达的改变,影响细胞应对不同机械力刺激时胞内信号传导能力,导致椎间盘组织胞外基质成分代谢和结构改变,最终诱发椎间盘突出的发生。对于机械力与细胞骨架的深入研究,可以进一步了解椎间盘突出的发病机制,为预防和治疗由椎间盘突出所引起的腰背痛和腰腿痛提供新的思路。
