不同剂量黄芪注射液对病毒性心肌炎小鼠Caspase-3活性的影响

摘要
目的观察黄芪注射液对病毒性心肌炎小鼠发生细胞凋亡及对半胱天冬氨酸蛋白酶3(Caspase-3)活性的影响。方法 46周龄雄性Balb/c小鼠60只,随机分为正常对照组、模型对照组、黄芪低剂量组、黄芪中剂量组、黄芪高剂量组5组,每组各12只。制备小鼠急性病毒性心肌炎模型后黄芪低、中、高剂量组分别腹腔注射黄芪注射液0.1、0.2、0.4mL/(g·d),正常对照组及模型对照组分别给予腹腔注射生理盐水0.1mL/(g·d),每日1次,连用14d。在第3、7、10、14天取心脏及血清标本,进行病理学检查、Caspase-3表达活性的检测、肌酸肌酶同工酶(CK-MB)及心肌肌钙蛋白Ⅰ(cTnⅠ)的检测。结果 (1)模型对照组小鼠心肌病理积分、CK-MB、cTnⅠ值显著高于正常对照组,差异有统计学意义(P<0.05);(2)黄芪低、中、高剂量组Caspase-3表达活性、CK-MB、cTnⅠ值、心肌病理积分均明显低于模型对照组,差异有统计学意义(P<0.05);(3)黄芪低、中、高剂量组各组间心肌病理积分、CK-MB及Caspase-3表达活性差异无统计学意义(P>0.05)。结论 (1)细胞凋亡参与了病毒性心肌炎发生时小鼠的心肌损伤,Caspase-3在其中起到重要作用;(2)小剂量的黄芪注射液即可以通过下调心肌细胞Caspase-3活性减少病毒性心肌炎小鼠心肌细胞凋亡,减轻心肌细胞损伤。
