生理药代动力学模型的特征及其国内外研究进展

摘要
介绍生理药代动力学(PBPK)模型的特征及其研究现状。通过Pubmed检索工具,查询国内外相关文献21篇,对PBPK模型的构建、模型特征和应用现状进行了文献整理和分析。目前PBPK模型已经广泛应用于有毒化合物的安全性评价、药物代谢过程研究、代谢酶和转运蛋白对药物代谢的影响、药物-药物相互作用以及新药的研发过程等研究之中。尽管PBPK模型有很多优势,获得了令人鼓舞的评价结果,但其构建需要数学和计算机的支持以及系统生物学、药物化学、药理学和数学、统计学等多学科专业人员合作,做进一步的探索和完善。
