异丙肾上腺素对大鼠心肌细胞凋亡相关基因表达的影响

摘要
目的 研究异丙肾上腺素 (isoprenaline,Iso)诱导大鼠急性心肌缺血与心肌细胞凋亡相关基因表达的关系。方法 sc大剂量Iso建立大鼠急性心肌缺血模型。RT PCR法检测凋亡相关基因Fas,CPP32 ,Bcl 2和Bax在心肌的表达。并用同样方法 ,观察苦豆碱 (aloperine,Alo ,2 0mg·kg-1)和普萘洛尔 (propranolol,Prop ,2mg·kg-1)对急性心肌缺血大鼠心肌细胞凋亡相关基因表达的影响。结果 急性心肌缺血模型大鼠心肌的Fas,CPP32和Bcl 2基因表达显著增加 ,Bax变化不明显 ;Alo和Prop能降低缺血心肌中Fas和CPP32基因的表达 ,增加Bcl 2基因的表达 ,且Prop对Bax表达有下调作用。结论 sc大剂量Iso可诱导大鼠心肌细胞凋亡相关基因异常表达 ;Alo和Prop对该模型大鼠心肌相应基因的异常表达亦有影响
