神经病理性痛大鼠脊髓NLRP3炎症小体表达的变化

摘要
目的评价神经病理性痛大鼠脊髓NOD样受体热蛋白结构域相关蛋白3(NLRP3)炎症小体表达的变化。方法健康成年雄性SD大鼠54只, 体重250~300 g, 采用随机数字表法将其分为2组(n=27):假手术组(S组)和神经病理性痛组(NP组)。S组仅暴露坐骨神经而不结扎;NP组采用坐骨神经慢性压榨性损伤法制备神经病理性痛模型。于术前1 d及术后1、4、7、14 d时测定机械缩足反应阈(MWT);于术后各时点MWT测定结束后处死, 取L4-6脊髓组织, 采用Western blot法检测NLRP3、凋亡相关的斑点样蛋白(ASC)、caspase-1、IL-1β的表达水平;采用实时荧光定量PCR法检测NLRP3、ASC、caspase-1、IL-1β的mRNA表达水平;于术后14 d MWT测定结束后取L4-6脊髓组织, 采用免疫荧光双标法检测NLRP3与胶质纤维酸性蛋白(GFAP)共表达情况。结果与S组比较, NP组术后各时点MWT降低, 脊髓NLPR3、ASC、caspase-1、IL-1β及其mRNA表达上调(P<0.05)。NLRP3和ASC及其mRNA表达水平于术后4 d时达峰值, caspase-1和IL-1β及其mRNA表达水平术后7 d时达峰值, MWT于术后7 d时达最低值(P<0.05)。NLRP3炎症小体主要表达于星形胶质细胞和神经元。结论外周神经损伤大鼠脊髓星形胶质细胞和神经元NLRP3炎症小体表达上调, 该变化可能参与了神经病理性痛的形成和维持。
