原花青素B2对人牙周膜细胞炎症介质表达的影响

摘要
目的·观察原花青素B2对牙龈卟啉单胞菌脂多糖(lipopolysaccharide,LPS)诱导的人牙周膜细胞(human periodontal ligament cells,hPDLCs)炎症介质表达的影响。方法·组织块法体外培养hPDLCs;噻唑蓝比色法观察原花青素B2对hPDLCs活性的影响;原花青素B2预处理hPDLCs 1 h后,予以LPS,实时荧光定量PCR和ELISA检测炎症因子白细胞介素1β(IL-1β)、IL-6及IL-8的mRNA和蛋白的表达;荧光显微镜下观察细胞内活性氧的变化;Griess法检测培养上清液中一氧化氮(NO)的水平。结果·当原花青素B2浓度达到100.00μg/mL时可增强hPDLCs的活性。原花青素B2可抑制LPS诱导的IL-1β、IL-6及IL-8的mRNA和蛋白的表达,降低活性氧和NO的水平。结论·原花青素B2可抑制牙龈卟啉单胞菌LPS诱导的hPDLCs炎症介质的表达,发挥一定的抗炎作用。
