FAK反义寡核苷酸促进人肺动脉平滑肌细胞凋亡

摘要
目的:了解黏着斑激酶(focal adhesion kinase FAK)是否参与人肺动脉平滑肌细胞(HPASMCs)凋亡。方法:纤黏连蛋白(FN40mg/L)刺激下培养的HPASMCs被用来进行反义寡核苷酸(antisense oligonucleotides,ASODNs)转染以做进一步实验。采用免疫印迹方法测定FAK蛋白质及凋亡蛋白半光氨酸蛋白酶-3(caspase-3)的表达,TUNEL法检测细胞凋亡。结果:免疫印迹方法测定结果表明,应用FAKASODNs转染后HPASMCs中FAK的表达明显低于转染前(P<0·01);caspase-3在应用FAKASODNs转染后高于转染前(P<0·05),TUNEL法检测结果表明FAKASODNs转染后细胞凋亡增加。结论:本实验结果表明FAKASODNs抑制细胞增生,促进细胞凋亡。其机制很可能是经caspase-3信号通路促进细胞凋亡。
