BAM22多肽对慢性炎性痛大鼠痛阈和电针镇痛作用的影响

摘要
目的采用大鼠右足底注射完全弗氏佐剂(complete freund's adjuvant,CFA)建立大鼠慢性炎性痛模型,观察牛肾上腺髓质22肽(bovine adrenal medulla 22,BAM22)对CFA致大鼠慢性炎性痛和电针(electroacupuncture,EA)镇痛作用的影响。方法将42只脊髓鞘内插管成功的健康雄性SD大鼠右足底注射CFA建立大鼠慢性炎性痛模型,造模后将大鼠随机分为生理盐水组、BAM22组、BAM22+纳洛酮组、生理盐水+EA组、BAM22抗体组和BAM22抗体+EA组,每组7只。生理盐水+EA组和BAM22抗体+EA组均在造模24 h后开始介入电针治疗,电针刺激大鼠双侧足三里、昆仑穴,刺激参数为疏密波,频率2/100 Hz,刺激强度1 mA,时间30 min,每日1次,连续7 d;余组仅以相应固定,不予电针处理。造模后第7天,各组大鼠鞘内注射相应试剂。分别检测造模前和造模后24 h、6 d、7 d时大鼠右足的热缩腿潜伏期(thermal paw withdrawal latency,T-PWL)作为痛阈指标。结果 BAM22多肽、电针组均可明显延长大鼠患侧T-PWL,鞘内注射纳洛酮能部分阻断BAM22多肽对大鼠痛阈的影响;BAM22抗体也可明显反转电针的T-PWL延长效应。结论 BAM22多肽和电针均能显著抑制CFA大鼠慢性炎性痛,电针镇痛作用可能存在与BAM22多肽相关的非阿片机制。
