基于NF-κB(p65)/CyclinE通路芪竹二术二草汤干预慢性萎缩性胃炎癌前期病变的研究

摘要
目的:观察芪竹二术二草方对萎缩性胃炎癌前病变的干预效果。方法:清洁级健康Wistar大鼠56只,随机分为阴性对照组(NC)、模型组、胃复春组(WFC)和仁术健胃颗粒组(RZ),每组10只,芪竹二术二草汤高剂量组(QZ-H)和低剂量(QZ-L)组,每组8只。模型组、WFC、RZ、QZ-H和QZ-L组采用化学诱癌物N-甲基-N’-硝基-N’-亚硝基胍(MNNG)诱发大鼠胃粘膜癌变,制备胃癌前期病变模型。WFC、RZ、QZ-H和QZ-L组大鼠分别按照0.39、3.2、54.4、13.6 g·kg-1剂量给予相应药物灌胃(ig),每天一次,连续28天;NC组和模型组大鼠给予等体积生理盐水。观察各组大鼠胃粘膜组织形态的改变,检测核转录因子(NF-κB,p65)和细胞周期蛋白E(CyclinE)的表达水平。结果:与阴性对照组相比,模型组大鼠炎细胞浸润、细胞异型增生程度及NF-κB/p65、CyclinE的表达明显增加,差异有统计学意义(P<0.01)。与模型组相比,WFC组大鼠NF-κB/p65及CyclinE表达减少,差异有统计学意义(P<0.05);RZ和QZ-L组大鼠NF-κB/p65及CyclinE表达明显减少,差异有统计学意义(P<0.01)。WFC、RZ、QZ-H及QZ-L组的复方总体有效率分别为50%、63.3%、43.3%及73.3%。结论:芪竹二术二草汤具有逆转萎缩性胃炎癌前期病变的作用,其作用机制可能是通过抑制原癌基因的转录和细胞周期调控蛋白的表达,从而防止肿瘤的发生发展。
