三七总皂苷对A549细胞侵袭转移的影响及机制研究

摘要
目的探讨三七总皂苷(PNS)对A549细胞侵袭转移的影响及机制。方法根据培养方法的不同将A549细胞分为对照组、PNS低剂量组(15 mg/L)、PNS中剂量组(30 mg/L)和PNS高剂量组(60 mg/L)。采用Transwell细胞侵袭实验检测A549细胞侵袭转移能力,q RT-PCR及Western blot法检测A549细胞mi RNA-379-5p、基质金属蛋白酶(MMP)-2及MMP-9的表达情况。结果与对照组比较,PNS低、中、高剂量组中A549细胞侵袭转移能力均下降,mi RNA-379-5p表达上调,MMP-2的m RNA及蛋白表达下调,差异均有统计学意义(P<0.05或P<0.01);PNS中、高剂量组中MMP-9 m RNA及蛋白表达下调(P<0.05)。结论 PNS可抑制A549细胞侵袭转移,其机制可能与PNS上调A549细胞mi RNA-379-5p表达和下调MMP-2、MMP-9表达有关。
