摘要
①目的 探讨紫外线B(UVB)对HaCaT细胞凋亡的影响及其作用机制。②方法 取培养的HaCaT细胞 ,用 30mJ/cm2 的UVB照射后继续培养 18h ,以流式细胞术检测细胞的凋亡率、线粒体膜电位和细胞内游离Ca2 +水平 ,用透射电镜观察细胞的超微结构。③结果 UVB照射增加了HaCaT细胞的凋亡率 (t =5 .2 36 ,P <0 .0 1) ,降低了线粒体膜电位 (t=6 .897,P <0 .0 1) ,升高了细胞内游离Ca2 + 水平 (t =6 .0 4 6 ,P <0 .0 1)。细胞的超微结构因UVB照射而遭到严重破坏 ,胞浆内的细胞器大量减少 ,线粒体严重空泡化 ,髓样小体形成 ,核内染色质成块并边聚。④结论 线粒体膜电位的下降和胞内游离Ca2 + 的升高可能参与了UVB诱发HaCaT细胞凋亡的过程
