PLA表位肽免疫微球诱导的CTL对肝癌细胞的杀伤作用

目的考察载肽聚乳酸微球(PLA)体外诱导特异性CTL杀伤活性。方法采用标准51Cr释放法检测CTL杀伤活性。结果免疫做球M1、M2在体外均能刺激人PBMC增殖,形成大量可见克隆;CTL杀伤分析结果显示,免疫微球M1、M2诱导的效应细胞对荷肽T2细胞(T2+P)、HepG-2和Alexander的杀伤率均达75%以上,二者的杀伤活性没有明显差异(P>0.05)。它们对表达hAFP的肝癌细胞HepG-2和Alexander的杀伤率均显著高于不表达hAFP的膀胱癌细胞BTT和未荷肽的T2细胞,差异明显(P<0.001)。结论免疫微球M1、M2均能在体外诱导产生特异性CTL,并对表达靶抗原的肿瘤细胞有较强杀伤作用。
