孕哺期接触多氯联苯对大鼠子代学习记忆及海马神经元的影响

摘要
目的探讨孕哺期接触多氯联苯对仔鼠学习记忆的影响及生理机制。方法成年Wistar大鼠受孕后用不同浓度的aroclor1254染毒, 仔鼠出生1个月后进行水迷宫实验, 分离海马细胞测定细胞内游离钙离子浓度, 透射电镜观察海马神经元的超微结构。结果各组仔鼠经过4 d训练后潜伏期与第1天相比均明显缩短(P<0.05), 但在同一天中染毒组与对照组的潜伏期差异无统计学意义;空间探索实验中, 各剂量组仔鼠第一次穿越平台时间与对照组相比均延长, 其中中、高剂量组与对照组差异明显[(11.74±6.56) s、(20.83±8.38)s, (5.23±1.16) s;均P<0.05)], 各剂量组在第四象限的停留时间与对照相比均明显减少[分别是(40.14±7.14) s, (33.76±5.58)s, (32.45±6.00)s, (30.63±5.10)s;P<0.05];穿越平台次数随着染毒剂量增加而减少[分别为(4.14±1.21)次, (3.00±1.32)次, (2.65±1.13)次, (2.42±1.31)次;P<0.05)], 中高剂量组差异具有统计学意义(P<0.05), 中高剂量组与对照组和低剂量组相比在目标象限停留时间明显减少[(40.14±7.14) s, (33.76±5.58) s, (32.45±6.00) s, (30.63±5.10) s);P<0.05]。染毒组细胞内游离钙离子浓度明显增高[分别为(121.16±12.23) nM, (141.27±24.66) nM, (163.32±29.75) nM, (261.46±27.79)nM], 中高剂量组差异具有统计学意义(P<0.05)。高剂量组海马神经元结构被破坏, 表现为线粒体肿胀, 脊排列紊乱, 核膜崩解, 脱髓鞘改变。结论 Aroclor1254孕哺期染毒能引起子代大鼠学习记忆能力降低和超微结构的改变, 这种作用可能与细胞内钙离子稳态失衡有关。
