跑台运动对慢性应激抑郁大鼠行为学及海马CA1、CA3区NGF表达的影响

摘要
为观察4周跑台运动对慢性不可预知性应激(CUS)致抑郁大鼠行为学、血浆促肾上腺皮质激素(ACTH)及皮质醇(CORT)含量、海马CA1、CA3区NGF表达的影响,将30只SD大鼠随机分为对照组、应激模型组及应激运动组,大鼠建立慢性不可预见性应激抑郁模型,同时应激运动组大鼠进行4周跑台运动。运动及应激结束后通过旷场实验、悬尾实验方法评定大鼠行为学指标变化情况,采用八臂迷宫实验测试大鼠的学习记忆能力,使用放射免疫法测试大鼠血浆ACTH及CORT含量,采用免疫组织化学结合图像半定量方法对海马CA1、CA3区NGF神经元的数量及面积进行测量和分析。结果显示:1)与应激模型组相比,应激运动组大鼠穿越格数、直立次数及修饰次数均显著增加(P<0.01,P<0.05,P<0.05),中央格停留时间显著缩短(P<0.05),悬尾不动时间缩短(P<0.05);2)与应激模型组相比,应激运动组大鼠完成八臂迷宫时间显著缩短(P<0.05),RME及TE均显著减少(P<0.05,P<0.05);3)与应激模型组相比,应激运动组大鼠血浆ACTH及CORT含量显著降低(P<0.05,P<0.01);4)与应激模型组相比,应激运动组大鼠海马CA1区、CA3区NGF免疫阳性细胞数量及面积均显著增加(P<0.05,P<0.05,P<0.05,P<0.05)。得出结论:跑台运动可改善抑郁模型大鼠的学习记忆能力,机理可能与长期运动降低抑郁大鼠的血浆ACTH及CORT水平、拮抗HPA轴功能亢进及增强海马CA1区、CA3区NGF的表达有关。
