半夏多糖对脂多糖诱导人肺微血管内皮细胞损伤的保护作用

摘要
目的探讨3种半夏(姜半夏、清半夏和法半夏)多糖(PSPR)对脂多糖(LPS)诱导的人肺微血管内皮细胞(h PMEC)炎症损伤的保护作用。方法以水提醇沉法提取PSPR;体外培养的h PMEC同时加入LPS 1 mg·L-1+PSPR 100和400 mg·L-1孵育24 h。CCK-8法测定细胞存活率,酶联免疫吸附试验(ELISA)测定肿瘤坏死因子α(TNF-α),白细胞介素1β(IL-1β),IL-6和IL-8释放,用Western蛋白质印迹法检测细胞IL-8和细胞间黏附分子1(ICAM-1)的表达,实时定量PCR技术分析IL-8和ICAM-1 m RNA水平。结果与溶剂对照组相比,LPS 1 mg·L-1损伤组细胞存活率显著降低(P<0.01);与LPS 1 mg·L-1损伤组相比,3种PSPR 100和400 mg·L-1能有效缓解LPS诱导的细胞存活率下降(P<0.01),抑制LPS诱导的TNF-α,IL-1β和IL-6释放(P<0.01),抑制LPS诱导的IL-8和ICAM-1蛋白表达增加(P<0.01)及IL-8和ICAM-1 m RNA水平增加(P<0.01)。结论 3种PSPR对LPS诱导的h PMEC炎症损伤有一定的保护作用,其机制可能与抑制TNF-α,IL-1β和IL-6等促炎因子的释放有关;PSPR还可抑制IL-8和ICAM-1的表达,有助于缓解IL-8和ICAM-1引起的中性粒细胞趋化作用。
