不同浓度脂多糖刺激下成骨细胞的增殖分化凋亡及IL-23表达

摘要
目的研究小鼠成骨细胞在不同浓度脂多糖(Lipopolysaccharides,LPS)刺激下的增殖分化凋亡及白介素-23(interleukin-23,IL-23)的表达。方法第一实验组至第五实验组分别用含0.01u g/ml、0.1u g/ml、1ug/ml、10ug/ml、100ug/ml LPS的培养基对小鼠成骨细胞MC3T3-E1进行刺激,并以不加LPS刺激的作为对照组,培养24小时后,应用MTT法、碱性磷酸酶(alkaline phosphatase,ALP)法检测六组细胞增殖、分化情况,流式细胞术(flow cytometry,FCM)和Western Blot检测细胞凋亡情况,并采用酶联免疫吸附剂(enzyme linked immunosorbent assay,ELISA)方法检测六组IL-23的蛋白表达。结果 LPS刺激下各实验组MTT和ALP检测值均低于对照组,差异有显著性(P<0.05),且随LPS浓度增加有逐渐降低趋势;LPS促进了MC3T3-E1的凋亡。实验组IL-23蛋白的表达均高于对照组,差异有显著性(P<0.05),且随LPS浓度增加有逐渐升高趋势。结论 LPS抑制成骨细胞增殖分化,刺激成骨细胞表达IL-23,且呈剂量依赖性。
