心肌细胞横管的形态发生:Junctophilin-2的作用

摘要
横管系统是心脏和肌肉细胞兴奋收缩偶联的结构基础.在新生鼠的心肌细胞中并不存在横管结构,横管的形态发生是心肌细胞成熟过程中的一个重要标志.本文对心肌细胞成熟过程中横管的形态变化进行定量分析,并对连接横管和肌质网的关键分子Junctophilin-2(JP2)在这个过程中所发挥的作用进行研究.对心肌细胞图像的分析显示,在横管成熟的过程中,横管的横向组分逐渐增加,纵向组分逐渐减少.通过对横管图像进行傅里叶变换分析,发现横管排列的规律性随心肌细胞的成熟而逐渐增加.横管的这些变化伴随着JP2蛋白表达量的上升,并且可以被JP2表达量的敲减所逆转.这些现象表明,JP2在横管形态发生过程中发挥重要作用.
