黑曲霉bgl基因的cDNA克隆及序列分析

摘要
通过一步法提取黑曲霉AS3.796的总RNA,利用依据其他黑曲霉β-葡萄糖苷酶(bgl)基因序列设计的一对特异性引物,采用RT-PCR方法扩增得到了该菌bgl基因cDNA片段。将目的片段与pGEM-T载体连接并转入大肠杆菌。序列测定结果表明该cDNA片段大小为2583bp,其基因序列与已公布的其它黑曲霉bgl基因的同源性最高可达99%。蛋白质分析结果表明该β-葡萄糖苷酶属于糖苷水解酶家族3成员。
