模式生物斑马鱼对川续断皂苷VI的代谢研究

摘要
首次采用模式生物斑马鱼研究续断中主要活性成分川续断皂苷VI的代谢,探索斑马鱼用于微量中药皂苷类成分代谢的可行性及合理性。采用高效液相色谱电喷雾质谱检测,Zorbax Extend C18(150 mm×4.6 mm,5μm)色谱柱,0.05%甲酸乙腈0.05%甲酸水梯度洗脱,正、负离子模式同时检测,根据准分子离子峰获得化合物分子量信息,通过与文献数据或对照品对照,鉴定川续断皂苷VI经斑马鱼暴露24 h后的药液及鱼体中的代谢产物。结果川续断皂苷VI经斑马鱼作用后,产生了脱去1分子、2分子及全部糖基的5种脱糖基代谢产物和1个羟基化产物,这与川续断皂苷VI在大鼠体内的代谢机制一致,提示斑马鱼对川续断皂苷VI的代谢具合理性,更具有化合物用量少,成本低、简单、高效的优势,特别是能体现在体代谢的综合结果,有望成为快速预测少量中药成分代谢、补充现有模型不足的全新的、模式生物代谢模型。
