高效液相色谱-串联质谱法测定青菜中雌酮、雌二醇和雌三醇残留

摘要
为了监测蔬菜被粪源类固醇激素污染状况,本试验建立同时测定青菜中雌三醇、17β-雌二醇、17α-雌二醇和雌酮残留量的高效液相色谱-串联质谱方法。青菜经乙腈超声提取,C18固相萃取柱去除杂质,氮吹浓缩处理后,用梯度洗脱上机检测。结果显示:青菜中雌三醇、17β-雌二醇、17α-雌二醇和雌酮得到很好的分离,标准曲线在10ng/ml至1 000 ng/ml浓度范围内呈线性关系,相关系数≥0.999,各自的保留时间分别为2.6 min、8.2 min、8.9 min和9.9min;当添加浓度为50μg/kg、100μg/kg和500μg/kg时,雌三醇、17β-雌二醇、17α-雌二醇和雌酮在青菜中的平均回收率分别为75.15%86.32%、73.82%83.21%、72.01%81.33%和81.08%90.35%,相对标准偏差为1.03%6.19%,该方法17β-雌二醇和17α-雌二醇的定量限均为10μg/kg,雌三醇的定量限为5μg/kg,雌酮的定量限为1μg/kg。表明该方法适用于蔬菜中雌三醇、17β-雌二醇、17α-雌二醇和雌酮残留的同步快速检测。
