鱼藤酮帕金森病模型的建立与评价

摘要
目的通过在颈背部皮下注射鱼藤酮试剂进行造模,建立帕金森病模型。方法将45只SPF级SD雄性大鼠随机分为正常组、假手术组、模型组,模型组给予颈背部皮下注射鱼藤酮试剂进行造模,假手术组仅注射等量葵花油乳化液(不含鱼藤酮),正常组不处理。观察其外观形态学改变,进行行为学测试、旷场实验、悬挂实验、滚轴实验并进行评分。免疫组织化学法检测大鼠黑质内α-突触核蛋白(α-synuclein,α-syn)与酪氨酸羟化酶(Tyrosine hydroxylase,TH)阳性表达。结果 (1)模型组大鼠出现弓背姿势,毛色变黄,背毛变硬呈扇形等改变。模型组大鼠行为学测试评分明显提高(P<0.01);旷场实验、悬挂实验、滚轴实验结果均显示模型组大鼠与正常组和假手术组相比行为异常(P<0.01)。(2)免疫组化检测结果显示模型组大鼠黑质内α-syn平均光密度值显著增高(P<0.01),TH平均光密度值显著降低(P<0.01)。结论用颈背部皮下注射鱼藤酮试剂的方法可建立与人类帕金森病行为和病理高度相似的大鼠模型,为PD实验的模型建立提供理论基础。
