α-银环蛇毒素的分离提纯和碘标记

本文介绍了α-银环蛇毒素的分离提纯、碘标记和鉴定。用羧甲基葡聚糖C-50离子交换层析和葡聚糖G-25层析,从湖南银环蛇毒液中制备出纯α-银环蛇毒素。均质α-银环蛇毒素是用羧甲基纤维素CM-32重层析纯α-银环蛇毒素（第3峰）而获得。用氯胶-T法进行125I标记α-银环蛇毒素。经Sephadex G-25柱层析分离纯化,产品放射性比度为90～100Ci/mM,标记率40～50％,蛋白回收率40～60％,游离碘含量小于3％。用测定N-末端氨基酸,氨基酸成分,等电点和聚丙烯酰胺凝胶电泳进行纯α-银环蛇毒素的鉴定。以小白鼠毒性,神经肌肉阻断作用和结合胆硷能受体等测定α-银环蛇毒素的生物活性。
