栀子苷通过ERK1/2-Nrf2通路发挥对Aβ25-35诱导的阿尔兹海默病大鼠模型的神经保护作用

摘要
目的探讨栀子苷对阿尔兹海默病(AD)大鼠模型的神经保护作用及机制。方法将60只大鼠随机分为5组:假手术组(Sham)、模型组(Model)、栀子苷小剂量组(12.5mg/kg)、中剂量组(25mg/kg)和大剂量组(50mg/kg),每组12只大鼠。模型组和不同剂量栀子苷组通过侧脑室微量注射Aβ25-35建立大鼠AD模型,假手术组注射等量生理盐水。大鼠AD模型建立后,栀子苷组以不同剂量栀子苷溶液灌胃,1次/d,连续灌胃给药8d;假手术组和模型组给予等量的生理盐水。药物治疗结束后,Morris水迷宫检测大鼠记忆能力和空间探索能力。大鼠处死,取出脑组织,HE染色和Highman刚果红染色评估海马区神经元损伤程度和淀粉样物质沉积情况,Western blot实验检测蛋白表达量。结果 Morris水迷宫实验显示,与假手术组比较,模型组潜伏期时间明显延长(P<0.05);与模型组比较,栀子苷组以剂量依赖性的方式缩短大鼠潜伏期,差异具有统计学意义(P<0.05)。空间探索实验测试结果显示,与假手术组比较,模型组大鼠穿越的平台次数和平台滞留时间明显降低(均P<0.05);与模型组比较,栀子苷组以剂量依赖性的方式增加大鼠穿越平台次数和平台滞留时间(均P<0.05)。模型组大鼠海马区神经元大量丢失,淀粉样物质沉积增多,栀子苷组剂量依赖性地改善神经元结构和减少淀粉样物质沉积。与假手术组比较,模型组Bax表达明显上调,Bcl-2表达及Bcl-2/Bax比例明显降低;与模型组比较,栀子苷组呈剂量依赖性地上调Bcl-2表达及Bcl-2/Bax比例,降低Bax蛋白表达(均P<0.05)。与假手术组比较,模型组p-ERK1/2、Nrf-2蛋白表达明显降低(均P<0.05);与模型组比较,栀子苷组呈剂量依赖性地上调p-ERK1/2、Nrf-2蛋白表达,组间比较差异均具有统计学意义(均P<0.05)。结论栀子苷对Aβ25-35诱导的神经元损伤具有保护作用,明显改善AD大鼠认知和行为能力,其作用机制可能与激活ERK1/2-Nrf2信号通路有关。
