大豆主栽品种体细胞胚胎发生的影响因素及再生植株

摘要
以大豆3个主栽品种3.0—6.0mm幼胚的子叶为外植体研究体外体胚发生的各影响因素。发现不仅激素种类、浓度及其组合而且幼胚长度、接种量等因素对体胚发生方式有重要影响。通过因素方差分析实验表明除幼胚长度、激素浓度、基本培养基种类对胚性反应频率有极其显著作用外,且发现前两种因素存在交互作用,并刚好达到极其显著水平(1%),最佳2.4-D浓度与幼胚长度组合为10D/4.0mm;20D/5.0mm;40D/5.0mm。在此组合下,基本培养基E1明显优于MS。总之,在我们建立的再生系统中,大豆未成熟子叶的体胚发生频率已达50%以上,由体胚正常萌发成植株的频率达52.9%—62.6%。
