桑椹及桑椹酒多酚提取物对BAT-cMyc细胞增殖及分化的影响

摘要
观察桑椹和桑椹酒多酚提取物对棕色脂肪细胞BAT-cMyc增殖的影响,探讨桑椹和桑椹酒多酚提取物对BAT-cMyc细胞棕色脂肪分化的效果。用不同浓度的桑椹和桑椹酒多酚提取物干预BAT-cMyc细胞48h。采用MTT比色法检测桑椹和桑椹酒多酚提取物对BAT-cMyc细胞的生长抑制作用,计算IC50,并实时荧光定量法测定细胞增殖相关基因Ki67及PCNA相对表达量。再通过桑椹和桑椹酒多酚提取物处理BAT-cMyc细胞,直至分化第6d,收集细胞。采用油红O染色法和实时荧光定量法对脂肪细胞分化效率进行分析。结果发现,桑椹和桑椹酒多酚提取物对BAT-cMyc细胞生长抑制的IC50浓度分别为186.37μg/mL和158.78μg/mL,且随着提取物浓度的升高对BAT-cMyc细胞的抑制增强。在浓度低于10μg/mL时,桑椹和桑椹酒多酚提取物对BAT-cMyc细胞的增殖无影响,却可以显著增加BAT-cMyc的脂肪细胞的分化和上调棕色脂肪细胞分化相关基因,如AP2和C/EBPδ等的表达。
