一种新的半月板切断导致小鼠骨关节炎方法的建立

摘要
目的建立一种创面小、操作简单且重复性高的半月板切断术诱发小鼠骨关节炎的手术方法。方法小鼠膝关节内侧皮肤剪开暴露术野,剪断内侧副韧带形成一个小切口,切断半月板上下缘的关节囊暴露内侧的半月板,从切口处进针,将内侧半月板用改造过的胰岛素注射笔针头钩出并剪断,缝合肌肉及皮肤。结果番红O染色结果表明,与对照侧关节相比,术后1周,全部手术侧关节表面开始发生磨损,关节软骨胞外基质蛋白聚糖减少;随着时间延长,手术侧关节表面磨损范围逐步扩大,关节软骨胞外基质蛋白聚糖丢失程度明显加剧,直至软骨下骨暴露于关节腔中。手术侧Mankin分值显著高于对照侧,且随术后时间延长手术侧Mankin分值逐步稳定递增。结论建立了一种新的半月板切断术,可简便、可靠地诱导小鼠发生骨关节炎。
