药物性肝损伤相关基因多态性研究现状

摘要
药物性肝损伤(drug-induced liver injury,DILI)指使用一种或多种药物后,由药物或其代谢产物引起的肝脏损伤,轻者停药后可自行恢复,重者可能危及生命。近年来对DILI危险因素研究较多,其中基因多态性成为热点。本文从DILI的流行病学、发病机制、临床分型以及药物基因组学几个方面进行综述,重点介绍了细胞色素P450单加氧酶(Cytochrome P450,CYP450)、尿核苷二磷酸葡萄糖醛酸转移酶(UDP-glucuronosyltransferase,UGT)、N-乙酰转移酶-2(N-acetyltransferase-2,NAT2)和人类白细胞抗原(human leukocyte antigen,HLA)的基因多态性和DILI的关系。
