单侧输尿管梗阻大鼠肾小管间质MCP-1的表达及其意义

摘要
目的 :探讨肾间质纤维化中单核细胞趋化因子 1(MCP 1)的表达及其与肾间质巨噬细胞浸润的关系。方法 :6 0只SD雌性大鼠随机分成假手术 (SOR)组、单侧输尿管梗阻 (UUO)组 ,于术后第 1,4 ,7,10 ,14d分别处死各组大鼠。用HE和MASSON染色法观察UUO大鼠术后不同时间点肾脏病理改变 ,用免疫组化法检测肾小管间质MCP 1表达和巨噬细胞浸润。结果 :UUO组肾小管 间质MCP 1,单核巨噬细胞抗原ED 1表达较SOR组明显增多 (P <0 .0 5 ) ,MCP 1表达与巨噬细胞浸润呈正相关 (r =0 .86 5 ,0 .92 8,0 .85 8,0 .893,0 .873,均P <0 .0 5 ) ;MCP 1表达和巨噬细胞浸润分别与肾间质胶原相对面积呈正相关 (分别为r =0 .85 6 ,0 .896 ,0 .6 86 ,0 .82 0 ,0 .86 7和r =0 .94 2 ,0 .898,0 .92 0 ,0 .914 ,0 .82 9,均P <0 .0 5 )。结论 :随UUO大鼠病程进展 ,肾小管间质MCP 1表达增多 ,可能在肾间质纤维化中起重要作用。MCP 1可能通过介导巨噬细胞浸润参与肾间质纤维化 ,MCP 1可成为治疗肾间质纤维化的新靶点
