肿瘤坏死因子-α抑制3T3-L1脂肪细胞色素上皮衍生因子的表达和分泌

摘要
目的:探讨肿瘤坏死因子-α(Tumor necrosis factor-α,TNF-α)对3T3-L1脂肪细胞色素上皮衍生因子(Pigment epithelium-derived factor,PEDF)表达和分泌的影响。方法:体外培养和诱导分化3T3-L1前脂肪细胞。分别用不同浓度TNF-α(0.1、1、10、100 ng/ml)处理3T3-L1脂肪细胞24 h,并选取10 ng/ml的TNF-α干预细胞3、6、12、24 h,采用实时荧光定量PCR和ELISA技术检测3T3-L1脂肪细胞PEDF的mRNA表达和蛋白分泌情况。结果:①和对照组相比,不同浓度TNF-α对3T3-L1脂肪细胞PEDF的mRNA表达和分泌均有抑制作用,且浓度越大,其抑制效应越明显(P<0.05)。②10 ng/ml的TNF-α干预3T3-L1脂肪细胞后,随着时间的延长,PEDF的mRNA表达逐渐降低,第12 h达到最低点(P<0.05)。③10 ng/ml的TNF-α对其PEDF分泌的抑制率在第6 h为15%,在第24 h为33%(P<0.05)。结论:TNF-α抑制3T3-L1脂肪细胞PEDF的表达和分泌,其抑制效应呈剂量和时间依赖性。
