诱导多能干细胞治疗大鼠糖尿病足溃疡的效果研究

摘要
目的观察局部移植异体诱导多能干细胞(induced pluripotent stem cell,IPSC)治疗糖尿病足溃疡SD大鼠的效果及其对外周血血管内皮生长因子(vascular endot growth factor,VEGF)的影响。方法取SD雄性大鼠40只,随机分为IPSC治疗组和对照组,分别对两组造糖尿病足溃疡模型。造模成功后IPSC治疗组在溃疡创面创缘肉膜层多点注射IPSC悬浊液,对照组在溃疡创面创缘肉膜层多点注射等体积Knockout DMEM培养液。观察比较两组治疗第1、5、10天溃疡面积及外周血VEGF水平。结果 40只SD雄性大鼠均成功造模。两组溃疡面积比较,治疗第1天差异无统计学意义(P>0.05);治疗第5、10天IPSC治疗组溃疡面积均小于对照组,差异有统计学意义(P<0.05)。两组外周血VEGF水平比较,治疗第1天差异无统计学意义(P>0.05);治疗第5、10天IPSC治疗组外周血VEGF水平均高于对照组,差异有统计学意义(P<0.05)。结论异体移植IPSC可促进糖尿病足溃疡SD大鼠溃疡愈合及局部新生血管形成。
