感染性脑损伤中血清高迁移率族蛋白B1的表达及丙酮酸乙酯的保护作用

目的探讨幼年大鼠感染性脑损伤时外周血中高迁移率族蛋白B1(High mobility group protein B1,HMGB1)的表达规律及丙酮酸乙酯(Ethyl pyruvate,EP)对其的保护作用。方法 150只1月龄SD大鼠随机分为对照组(NS组,50例),颈外动脉注射生理盐水;感染性脑损伤组(LPS组,50例),颈外动脉注射LPS;EP治疗组(EP组,50例),颈外动脉注射LPS后即刻腹腔注射EP,于注射药物后6、12、24、48、72 h取外周静脉血,采用酶联免疫吸附试验分析HMGB1、神经元烯醇化酶(Neuron specific enolase,NSE)水平。结果 LPS组外周血HMGB1、NSE水平显著增高,各时间点与NS组比较差异有统计学意义。EP干预后外周血HMGB1及NSE水平均显著下降(P均<0.05),LPS组及EP组HMGB1与NSE水平均呈正相关。结论 HMGB1可能作为晚期炎症因子参与感染性脑损伤的发生发展过程,外周血HMGB1的检测可较好地反映感染性脑损伤时HMGB1的表达规律;丙酮酸乙酯可能通过抑制HMGB1的表达而对感染性脑损伤具有保护作用。
