抑制水通道蛋白1对Hep-2细胞增殖和凋亡的影响

摘要
目的:观察水通道蛋白1(AQP-1)对喉癌Hep-2细胞增殖和凋亡的影响。方法:应用MTT法、流式细胞仪及原位末端标记(TUNEL)法、免疫组织化学SABC方法多种技术分别检测AQP-1的抑制剂乙酰唑胺对细胞生长抑制率、凋亡率和细胞内AQP-1蛋白表达的影响。结果:乙酰唑胺在5×10-5mol/L浓度下对Hep-2细胞内AQP-1蛋白表达有明显的抑制作用,细胞生长抑制率和细胞凋亡率均随时间延长而增大,呈时间依赖性。并且TUNEL法检测的细胞凋亡率与免疫组织化学SABC方法检测的AQP-1蛋白抑制率呈明显正相关(r=0.784,P<0.05)。结论:乙酰唑胺在一定浓度下对AQP-1的抑制可显著诱导喉癌Hep-2细胞凋亡,提示AQP-1可能成为治疗喉癌新的靶点。
