血管内皮生长因子在动脉粥样硬化缺血性脑卒中模型大鼠脑组织中的表达

摘要
目的探讨动脉粥样硬化基础上缺血性脑卒中模型大鼠脑组织中血管内皮生长因子表达规律及意义。方法大鼠被随机分为对照组和模型组,后者制备动脉粥样硬化大鼠模型,然后再被分为动脉粥样硬化模型假手术组、动脉粥样硬化模型持续缺血0.5、6、12、24 h组,运用逆转录聚合酶链式反应技术检测血管内皮生长因子mR-NA表达及免疫组织化学法检测血管内皮生长因子蛋白表达及变化。结果各动脉粥样硬化模型组与对照组比较血管内皮生长因子表达有明显升高(P<0.05),各持续缺血组与假手术组比较血管内皮生长因子基因及蛋白表达均有明显升高(P<0.05),并随时间发生规律性变化,其中血管内皮生长因子的基因及其蛋白产物在持续缺血6 h组均达到峰值。结论机体长期存在动脉粥样硬化基础病变对缺血性脑卒中脑组织血管内皮生长因子基因及其蛋白产物的表达有正向调控作用,缺血后可随时间变化。本实验结果可为动脉粥样硬化基础上缺血性脑卒中的发生、发展、治疗及预后估计提供一定的理论基础。
