人MAPK3基因克隆及其在肝细胞癌细胞中的作用

摘要
目的 克隆人MAPK3基因,探讨MAPK3基因对肝细胞癌细胞的作用。方法 以肝细胞总RNA为模板,利用RT-PCR体外扩增获得人MAPK3基因的蛋白编码区,并构建重组真核表达载体pEYFP-MAPK3;脂质体转染和G418药物筛选获得MAPK3稳转肝癌细胞株;蛋白免疫印迹检测稳转肝癌细胞株中外源MAPK3蛋白的表达量;实时无标记细胞分析技术检测MAPK3基因对肝癌细胞增殖的影响;划痕愈合实验和Transwell侵袭实验分别MAPK3基因对肝癌细胞迁移和侵袭的影响;蛋白免疫印迹检测ERK信号通路靶基因及上皮-间质细胞转化标志物的表达变化。结果 RT-PCR克隆获得人MAPK3基因,成功构建重组真核表达载体pEYFP-MAPK3并获得MAPK3基因稳转肝癌细胞株,发现MAPK3基因可以促进人肝细胞癌细胞的增殖、迁移和侵袭,通过磷酸化活化Stat3,促进c-Myc、MMP-2和N-Cadherin表达升高和抑制E-Cadherin表达。结论 成功克隆人MAPK3基因并证明MAPK3可以有效促进肝细胞癌细胞的增殖、迁移和侵袭。
