荧光原位杂交在喀斯特山地土壤硫酸盐还原菌检测中的应用

摘要
采用荧光原位杂交(Florescence in situ Hybridization,FISH)技术分析了喀斯特山地土壤硫酸盐还原菌(SRB)的数量和空间分布状况。运用16SrRNA特异性探针,结合DAPI全细胞染色和样品稀释,FISH技术准确、高效地原位检测出土壤剖面中SRB的数量和分布状况。结果显示,在所研究的土壤剖面各层均有SRB检出,最低为(0.8±0.4)×107个/g,最高为(6.2±1.3)×107个/g,平均为(2.7±1.2)×107个/g。FISH能同时对土壤中的SRB进行定性和定量分析,是研究环境中SRB时空分布的快速有效的检测技术。
