淫羊藿苷通过上调VEGF信号通路促进脑低灌注大鼠的血管发生(英文)

摘要
目的观察淫羊藿苷对大鼠慢性大脑皮层低灌注后血管新生的作用。方法采用双侧颈总动脉结扎(BCCAO)制备大鼠脑低灌注模型,不结扎大鼠为假手术对照。术后9 d分组:假手术组、淫羊藿苷-60、BCCAO、BCCAO+淫羊藿苷-15、BCCAO+淫羊藿苷-30和BCCAO+淫羊藿苷-60组。各组大鼠灌胃相应剂量的淫羊藿苷或蒸馏水,连续28 d。采用免疫荧光染色,CD31抗体标记毛细血管密度;采用Griess方法检测大脑皮层一氧化氮(NO)的含量;采用Western blot方法检测血管内皮生长因子(VEGF)信号通路关键分子的蛋白表达水平及磷酸化程度。结果 BCCAO大鼠脑内CD31阳性标记的血管数量减少;大脑皮层NO浓度降低;VEGF信号通路下调。而给予淫羊藿苷治疗28 d后,能部分逆转上述改变,表现为增加缺血脑区毛细血管数量,提高大脑皮层NO浓度,上调VEGF信号通路。结论淫羊藿苷对慢性脑低灌注时血管新生具有积极的促进作用,该作用与增加受损大脑皮层NO浓度和上调VEGF信号通路有关。
