胡黄连苷Ⅱ通过激活自噬作用抑制泡沫细胞的形成与炎症反应

摘要
目的:从巨噬细胞泡沫化角度探讨胡黄连苷Ⅱ抗动脉粥样硬化的作用机制。方法:CCK-8法确定胡黄连苷Ⅱ对细胞无毒性的浓度;氧化低密度脂蛋白(ox-LDL)诱导与药物干预后,采用油红O染色观察泡沫细胞的形成,酶法检测细胞内胆固醇水平;激光共聚焦显微镜观察胡黄连苷Ⅱ对巨噬细胞自噬的影响。采用Western blot检测巨噬细胞ABCA1、ABCG1、NLRP3、ASC的蛋白表达水平。结果:胡黄连苷Ⅱ对ox-LDL诱导的THP-1源巨噬细胞在0~100μmol/L的范围内无细胞毒性。各浓度胡黄连苷Ⅱ的干预均可抑制ox-LDL诱导的泡沫细胞形成。胡黄连苷Ⅱ浓度依赖性降低细胞内总胆固醇、胆固醇酯和胆固醇酯化率,显著增加ABCA1/ABCG1、LC3Ⅱ/Ⅰ和beclin1的表达;减少TNF-α、IL-1β和IL-6等炎症因子的表达,显著降低NLRP3、ASC和caspase-1的表达。结论:胡黄连苷Ⅱ显著抑制巨噬细胞泡沫化,相关的作用机制可能是通过促进巨噬细胞自噬,上调巨噬细胞ABCA1/ABCG1表达,抑制NLRP3炎症小体的激活,从而促进巨噬细胞内胆固醇外排,抑制炎症反应。
