柚皮苷对脂多糖刺激下成骨细胞碱性磷酸酶活性、骨保护素mRNA和RANKL mRNA的影响

摘要:
目的探讨不同浓度柚皮苷对脂多糖(LPS)刺激下成骨细胞碱性磷酸酶(ALP)活性、骨保护素(OPG)mRNA和核因子κB受体活化因子配体(RANKL)mRNA表达水平的影响。方法将小鼠胚胎成骨细胞前体细胞(MC3T3-E1)随机分为5组。空白对照组:加入等量的二甲基亚砜,未做其他处理。LPS组:加入终浓度为1 mg·L-1的LPS。低浓度组、中浓度组、高浓度组:在1 mg·L-1的LPS的基础上分别加入终浓度为2、10、20 mg·L-1的柚皮苷。24 h后采用CCK-8法检测成骨细胞增殖情况。3 d后使用碱性磷酸酶试剂盒检测成骨细胞ALP活性。采用逆转录聚合酶联反应(RT-PCR)检测OPG mRNA和RANKL mRNA表达水平。结果与空白对照组比较,LPS组成骨细胞增殖能力、ALP活性和OPG mRNA表达水平均较低,RANKL mRNA表达水平较高(P<0.05)。低浓度组、中浓度组、高浓度组的成骨细胞增殖能力、ALP活性和OPG mRNA表达水平均高于LPS组,低浓度组、中浓度组、高浓度组的RANKL mRNA表达水平均低于LPS组(P<0.05)。低浓度组成骨细胞增殖能力、ALP活性和OPG mRNA表达水平高于中浓度组和高浓度组,中浓度组成骨细胞增殖能力、ALP活性和OPG mRNA表达水平高于高浓度组(P<0.05)。低浓度组成骨细胞RNAKL mRNA表达水平低于中浓度组和高浓度组,中浓度组RNAKL mRNA表达水平低于高浓度组(P<0.05)。结论柚皮苷可降低LPS刺激下成骨细胞RANKL mRNA水平,上调OPG mRNA水平,进而抑制骨吸收,并且当柚皮苷的浓度为2 mg·L-1时的效果最显著。
