徒手切片与植物细胞微管免疫荧光定位

摘要
报道一种植物细胞微管骨架免疫荧光定位的新方法。化学固定后,通过徒手切片,将南瓜茎沿纵切面切成100~200μm的薄片,经过免疫荧光定位方法处理,在共焦激光扫描显微镜下,可以对不同类型细胞中微管骨架的排列进行观察。研究发现,南瓜茎细胞内微管骨架排列的主要方式有以下几种:1、网络状微管,多存在于薄壁细胞中,其中微管往往与液泡膜、核膜及质膜相连;2、周质微管,多存在于导管分子或伸长细胞中,其中微管往往彼此平行,并与细胞的纵轴成一定角度;3、纵向微管,存在于筛分子等少数伸长细胞中,其中微管往往接近与细胞纵轴平行。
