二甲双胍对多囊卵巢综合征大鼠的排卵及对卵巢中IL-6、TNF-α表达的影响

摘要
目的探讨二甲双胍对多囊卵巢综合征(PCOS)大鼠的促排卵作用及其对卵巢组织IL-6mRNA、TNF-αmRNA表达的影响。方法将32只雌性SD大鼠随机分为3组:空白对照组(A组,n=10),自由摄取食物和水31d;PCOS组(B组,n=12),来曲唑1mg/kg灌胃,21d后予以5mL/kg生理盐水灌胃10d;二甲双胍干预组(C组,n=10),来曲唑1mg/kg灌胃,21d后予以6%的二甲双胍5mL/kg灌胃10d。每日阴道涂片观察大鼠动情周期,末次灌胃24h后测定各组大鼠血清黄体生成素(LH)、雌二醇(E2)、睾酮(T)、胰岛素(INS)、空腹血糖水平,通过H-E染色观察卵巢形态,实时荧光定量PCR检测各组大鼠卵巢白细胞介素-6(IL-6)mRNA、肿瘤坏死因子-α(TNF-α)mRNA的相对表达。结果 C组较B组大鼠血清T、LH、INS明显降低(P<0.05),卵巢局部IL-6mRNA、TNF-αmRNA的表达下降(P<0.05),卵巢形态恢复正常。C组大鼠于用药的第6d全组恢复排卵,B组大鼠持续无排卵直至第10d,仅1只大鼠自发恢复排卵。结论二甲双胍能抑制卵巢局部IL-6mRNA、TNF-αmRNA的表达,有助于恢复卵巢形态,促进排卵,改善PCOS大鼠内分泌代谢紊乱。
