超声辅助双水相体系提取苦荞籽黄酮

摘要
为提高苦荞黄酮分离纯化效率,本文采用双水相体系超声辅助提取苦荞籽黄酮。本文探讨了15种有机试剂/无机盐双水相体系萃取苦荞黄酮的能力,并考察了液料比、超声时间、超声温度、硫酸铵和丙酮质量浓度在丙酮/硫酸铵双水相体系对黄酮分配行为的影响,同时采用响应面优化萃取条件。结果表明:15种有机试剂/无机盐双水相体系中,丙酮/硫酸铵双水相体系具有最佳的苦荞黄酮提取能力;苦荞黄酮最佳提取条件为:液料比46.65 g/g,超声温度44.75℃,硫酸铵质量浓度22.86%,在此条件下,黄酮得率可达20.01±0.26 mg/g;与80%乙醇提取法相比,本法所得黄酮提取物总黄酮含量和芦丁含量没有显著性差异,但黄酮纯度高达62.35%,纯度提高了38.86%;提取物DPPH·和ABTS+·清除率分别提高了22.52%、43.82%。因此,本法是一种更经济快速的苦荞黄酮初步提取纯化方法,有效提高了黄酮功能性整体研究进度。
