摘要
转化生长因子β(TGFβ)作用复杂,广泛参与哺乳动物的各种病理生理过程,影响细胞的增殖分化,与肿瘤的发生、发展密切相关。TGFβ信号通路关键的信号传导分子为胞浆蛋白Smads。TGFβ在正常细胞癌变初期可作为抑癌因子抑制细胞增殖,启动细胞分化或凋亡,而在肿瘤发展期则可因某些原因丧失对TGFβ应答的敏感性,刺激血管生成、抑制免疫反应、促进ECM形成,为肿瘤细胞的快速生长、转移提供良好的局部环境,反而成为促进因子,扮演着肿瘤抑制基因和癌基因的双重角色。文章综述TGFβ-Smads信号转导通路与肿瘤的相关信息。
