黄芪汤对糖尿病肾病小鼠AMPK-Camkkβ(LKB1)信号通路的影响

摘要
目的:探讨黄芪汤改善糖尿病肾病的作用及可能的机制。方法:40只6周龄db/db小鼠随机分为模型组及黄芪汤高、中、低剂量组,每组10只,10只6周龄db/m小鼠作为正常对照组。模型组、正常组均予0.5%羧甲基纤维素钠灌胃;各药物组分别予黄芪汤1.08 g/kg、0.36 g/kg、0.12 g/kg灌胃。1次/d,灌胃14周后进行生化指标、肾组织病理学检测,Western blot检测各组肾皮质中AMPK-Camkkβ(LKB1)信号通路相关蛋白的表达量。结果:与模型组比较,黄芪汤能降低db/db小鼠的体质量、肾质量、血清三酰甘油(TG)、胆固醇(TC)、低密度脂蛋白(LDL-C)浓度,降低血清谷氨酸转氨酶(ALT)、谷草转氨酶(AST)浓度,升高肾质量/体质量指数(P<0.05),改善肾脏病理情况;上调肾皮质中p-AMPK、p-Camkkβ、p-LKB1、p-ACC蛋白表达量(P<0.05)。结论:黄芪汤通过有效活化AMPK-Camkkβ(LKB1)信号通路改善糖尿病肾病。
